In April 2026 the FDA approved orforglipron, a small-molecule, non-peptide agonist of the GLP-1 receptor taken by mouth. It follows the late-2025 approval of an oral peptide formulation of semaglutide for weight management. [1]
A June 2026 pipeline review places approved and late-stage agents on a rough scale of weight reduction: orforglipron around 12 %, CagriSema (semaglutide with the amylin analogue cagrilintide, under FDA review) around 20 %, amycretin (one molecule acting on GLP-1 and amylin receptors, in Phase 3 from 2026) around 22 % in Phase 2, and retatrutide around 28 %. Further back sits a monthly antibody–peptide conjugate with Phase 2 results of 16 to 20 %. [1]
Oral agents generally produce less weight reduction than the leading injectables, and the figures above come from different trials with different populations; they are not head-to-head results. [1]
