What was done
Retatrutide is an investigational once-weekly agonist of the GIP, GLP-1 and glucagon receptors. TRANSCEND-T2D-1 (type 2 diabetes, 40 weeks) reported on 19 March 2026; TRIUMPH-1 (obesity, 2,339 participants, 80 weeks with a 104-week extension) on 21 May; TRIUMPH-2 (obesity with type 2 diabetes) and TRIUMPH-3 (obesity with cardiovascular disease) on 23 July. [1][2][3]
What was found
In TRANSCEND-T2D-1 the 12 mg dose lowered A1C by 1.7 to 2.0 points and reduced body weight by 16.8 % at 40 weeks. In TRIUMPH-1, mean weight reduction at 80 weeks was 19.0 % (4 mg), 25.9 % (9 mg) and 28.3 % (12 mg) against 2.2 % on placebo; participants escalated to the highest tolerated dose reached 30.3 % at 104 weeks. TRIUMPH-2 reported 20.8 % and TRIUMPH-3 22.6 % at 80 weeks on 12 mg. [1][2][3][4]
Limits
All four are sponsor press releases, not peer-reviewed papers. Adverse events were described as consistent with the incretin class, with gastrointestinal effects predominant; discontinuation by dose is not fully reported in the releases. The cardiovascular outcomes trial is ongoing. [2][4]
Funding
Eli Lilly and Company, the developer, sponsored all four trials and issued the results. A regulatory submission is planned for the first quarter of 2027. [3]
